Accumulation of TCR signaling from self-antigens in naive CD8 T cells mitigates early responsiveness https://www.biorxiv.org/content/10.1101/2023.01.27.525946v1?med=mas
'Immune response to the vaccine mRNA is the most literal example of “killing the messenger” that I can think of, short of the Spartan treatment of Darius’ envoys in 491 BC.'

Plenty of good vaccine news lately, unfortunately not regarding HIV: J&J just declared to discontinue their phase 3 HIV vaccine trial with a mosaic-based Ad26.

My first post on Mastodon after a whole lot of lurking and boosting, so I'm treating this as a mini #introduction for myself

I'm an #immunology PhD candidate at #ColumbiaUniversity medical center, and we just published a study of #TissueSpecifity of human #Tcell identity and clonal expansion in barrier sites, using #CyTOF, #scRNAseq, and #TCR #sequencing!

Tissue adaptation and clonal segregation of human memory T cells in barrier sites - Nature Immunology

Farber and colleagues examine the phenotypic, transcriptomic, clonal, and functional differences between tissue-resident T cells in various barrier tissue sites relative to T cells in lymphoid organs and circulation in humans.

#Influenza A, which started circulating at high levels earlier this season than in any season since the 2009 #H1N1 pandemic, is declining as quickly as it climbed. It is truly remarkable to see the map of #flu-like activity show so little red in the 2nd week of Jan. It is like this year's flu season shifted forward 2 months or so.
Last week on the right. Same week in 2018, on the left, as the 2017-18 season started to take off.
@victoralab @jbloom_lab Loved the preprint already. Highly relevant and elegant at the same time. Congrats again.

Happy to see this study out! Massive effort by our student Arien Schiepers and also many collaborators, both from our lab and @jbloom_lab, Paul Bieniasz (Rockefeller), and Norbert Pardi’s (U. Penn) groups.

Molecular fate-mapping of serum antibody responses to repeat immunization - Nature

Serum antibody responses to sequential homologous booster vaccines derive overwhelmingly from primary cohort B cells at the expense of de novo responses; this ‘primary addiction’ can be overcome by boosting with variant antigens.

If you might be interested in reading some nuanced views on antigenic imprinting, please see this from @katherinejwu https://www.theatlantic.com/health/archive/2023/01/covid-vaccine-immunity-variant-protection/672704/. A better and more thoughtful read (with a lot of quotes from people who actually study this problem) than a certain NEJM commentary heavy on motivated reasoning.

Hi @Quartzy - is it true that from July onwards you will be charging also academic labs for using your basic order tracking system ($79/5 users)? Do I remember your #freeforever pledge correctly? Thanks for clarifying. #AcademicChatter
