A generalized framework for performance-based earthquake engineering: integrated assessment of structural reliability and resilience

Assessing structural performance under seismic hazard requires accounting for both damage accumulation and post-event recovery. In current performance-based earthquake engineering (PBEE), recovery is generally treated as a post-processing attribute, while structural performance is modeled using Poissonian exceedance assumptions that imply renewability and memorylessness. These assumptions hinder a unified treatment of reliability and resilience under repeated seismic loading. This study proposes a generalized PBEE framework in which damage and recovery are embedded directly into the system dynamics through a continuous-time Markov chain. A single generator matrix governs state-dependent transitions, providing a unified description of structural reliability and resilience while remaining compatible with standard PBEE metrics. Time-dependent failure probabilities and reliability indices are derived from the transient system dynamics, whereas resilience is quantified through the expected fraction of operational time before collapse. The framework exploits the spectral properties of the generator matrix to compute both metrics efficiently and transparently. The methodology is illustrated on a three-state example and applied to two structural archetypes: a braced frame and a base-isolated system. Results show that recovery dynamics can strongly affect long-term resilience even when conventional reliability measures exhibit limited sensitivity, emphasizing the need to explicitly account for recovery in life-cycle seismic performance assessment.

Resistance to first-line osimertinib in EGFR-mutant non-small-cell lung cancer (NSCLC) is the canonical example of predictable clonal evolution under therapeutic pressure, yet no public benchmark exists for training or evaluating computational models on the corresponding longitudinal patient trajectories. We introduce OncoTraj, a public benchmark of 813 EGFR-mutant NSCLC patients receiving first-line osimertinib, harmonized from three real-world clinical-genomic sources: MSK-CHORD (672 patients), AACR Project GENIE BPC NSCLC (34 patients), and the FLAURA molecular-resistance supplement (107 patients). OncoTraj defines three locked tasks: (A) binary classification of progression by a fixed 12-month landmark, (B) regression of time-to-first-progression in days, and (C) six-class classification of the dominant resistance mechanism. We release the harmonized dataset, patient-level train/validation/test splits with an audited no-leakage guarantee, an open-source evaluation harness, and six reference baselines spanning a majority-class predictor, logistic regression, random forest, XGBoost, an LSTM, and a multi-task transformer. With v1's single-timepoint snapshot features, no task clears chance on clean within-source evaluation: the uniformity of this ceiling across every model class localizes the limit to the input modality (single-snapshot tissue NGS rather than serial ctDNA), not the algorithm. The benchmark does recover a reproducible literature-consistent association: TP53 co-mutation raises the 12-month progression rate from 29% to 59% cohort-wide. OncoTraj establishes a reproducible, leakage-audited baseline and converts the modality limit into concrete design requirements for a serial-ctDNA-enriched v2.

Multimarginal flow matching with optimal transport potentials

Flow matching (FM) has emerged as a powerful framework for learning dynamic transport maps between two empirical distributions. However, less explored is the setting with intermediate observed marginals that can help constrain the flows between the endpoints. This "multimarginal" regime is central to modeling temporal evolution in dynamical systems in many scientific domains that can sample sequential distributions. We tackle this problem with a novel approach that leverages the connection between FM and dynamic optimal transport (OT), softly steering the flow towards the intermediate marginals through potential terms in the dynamic OT action. By extending the conditional FM learning target to incorporate these potentials, we derive an efficient, simulation-free algorithm for multimarginal FM that offers considerable flexibility in the spatiotemporal dynamics of the learned flows. We demonstrate state-of-the-art performance and training efficiency of OT-potential FM (OTP-FM) on diverse single-cell RNA sequencing, oceanographic, and meteorological datasets.
