RNA-triggered cell killing with CRISPR-Cas12a2

최근 발견된 CRISPR-Cas12a2는 특정 RNA 인식 시 무차별적인 이중가닥 DNA 절단 활성을 발휘하여 효모와 인간 세포에서 목표 RNA를 발현하는 세포를 선택적으로 제거할 수 있다. Cas12a2는 표적 전사체에 의해 활성화되며, 오프타겟 활성화 없이 세포 사멸을 유도한다. 이를 활용해 인유두종바이러스 감염 세포, 유전자 편집 실패 세포, KRAS의 종양 유발 점 돌연변이 세포를 선택적으로 제거하는 데 성공했다. 기존 Cas9, Cas12a, Cas13 기반 방법들이 진핵세포에서 세포 사멸을 효과적으로 유도하지 못한 한계를 극복하는 새로운 CRISPR 도구로 주목된다.
