Haojia WuWhen no mRNA is expressed from an enhancer in given a cell type, how to directly measure the reporter expression? Single-cell quantitative expression reporters (scQers) developed by Shendure lab can solve this problem. Key designs: 1) A dual RNA cassette decouples the detection and quantification (oBC for detection and mBC for quantification). 2) oBC are circularized Pol III transcribed barcodes which expressed >150-fold higher steady-state levels. #singlecell #devbio

So genomic.social has been active for 20 days. How are you finding it? does anyone have any feedback?
Once we have been up for a month I'll give some more detailed cost breakdowns and detailed information about the instance. I'll share my rough modelling for the future too (as it will be a tad clearer with a month of data)

For what % of the proteins in a cell line, if you tag their endogenous locus with a GFP (one allele), the GFP levels will be high enough to be seen/sorted by FACS? Top 100 most abundant? 1K? 5K? Can extrapolate from FPKM?
