I've never done a PVP night and I doubt I ever would. Too easy to get hurt feelings, even if it's just a one-off, not really impacting continuity kind of thing.

I'm sure it would work well for a certain kind of group and I'm glad they're enjoying it.
As far as scenarios go, I could see a "contest of champions" for bragging rights or maybe some interesting in-game gear (maybe nothing unbalancing towards combat but some fund magic items could be interesting; maybe the winner could pick and the second place gets second pick, etc.).
@cynical13 I've had prizes before, things like dice bags or trays, etc. Coming up with scenarios that don't give unfair advantages to certain characters is the biggest challenge. We've yet to have personality or hard-feelings issues come up as they all know it's in-game and not personal (and deaths don't count toward the ongoing campaign).
One issue I can see is that classes are usually designed for a particular play style. You'd almost have to have a striker league and a controller league or even just Rogues and just Wizards or Clerics. Otherwise, I could see it becoming more of a rock/paper/scissor kind of event.
