@siderea in genetics speak, a mutation can be gain of function or loss of function. Loss of function is also just what it sounds like, a mutation that reduces or eliminates the function of a gene. Since this mutation results in reduced expression of a transcriptional target, they're emphasizing here that the reduced transcription is actually due to a gain of function in a transcriptional regulator
