New publication from the lab!
We coevolved a phage-producing + phage-sensitive strain and tested the different phage resistance mechanisms, their emergence and their dynamics.
Unexpectedly, new lysogens are unstable and undergo excessive death rate during expo growth.
Other mechanisms of resistance like receptor (capsule) loss are fitter in short evolution times, but later, clones with receptor modifications or displaying phenotypic resistance are fitter.

We speculate that when the capsule is added (exogenously) to a culture, it can bind antimicrobial peptides (AMPs).
Such sequestration titers AMPs in the media, effectively diluting it & ⤴️⤴️ tolerance, as more compound is required to obtain the same killing effect.

But when the capsule is bound to the cell surface (native form), it facilitates the entry of the AMPs as it retains AMPs near the cell surface where it can easily bind to the LPS, & ⤴️⤴️ susceptibility.

We tested the MIC, growth @ sub-MIC [] & death rate of all non-capsulated mutants against 6 antibiotics including polymyxin B and last-resort colistin.

*The capsule certainly did not protect against antibiotics*

In fact, it REDUCED the MIC when cells were treated with polymyxins
