Did Gibson assembly cloning last night. Dropped four pieces into one plasmid backbone. A lot of colonies this morning. Worked better than I thought. In my experience, when I saw this many colonies, usually most of them are the right thing.

Last week I had a conversation with the boss about funding that I found eye opening. She would include in the project aspects that she is not actually interested but would likely increase the odds of it getting funded based on experience of what have been funded.
