Ohio State University oncologists found that certain non-small cell lung cancers (NSCLC) can survive treatment by activating a metabolic “backup system.” When one growth pathway is blocked, tumors increase glucose and fat metabolism to stay alive. The OSU researcher discovered that hitting two targets at the same time foils this survival system. The targets are: the lysosomes that process cell nutrients; and SREBP-1, a protein that helps cancer cells absorb glucose and build fats. When both pathways are blocked in test tube and animal studies, tumor cells showed major metabolic stress and cell death. One promising aspect is that several drugs used in this strategy already exist could help speed future clinical testing. They include chloroquine and simvastatin. This finding is key since some lung cancers don’t respond to immunotherapy and lack targetable mutations, leaving patients with limited options.
