Cancer biologists at the Oregon Health & Science University have developed a blood test, dubbed the PAC-MANN, that detects this form of pancreatic cancer with an 85% accuracy for early-stage cases. The test quantitates changes in protease activity that accompany development and progression of this cancer and is also useful for monitoring treatment effectiveness and cancer recurrences. https://www.science.org/doi/10.1126/scitranslmed.adq3110 #pancreatic #cancer #protease #pacmann".
Due to the limited efficacy of vaccines against SARS-CoV-2 and resistance to current therapies additional anti-viral therapeutics with pan-coronavirus activity are of high interest. Here, the authors screened 2.8 billion compounds from a DNA-encoded chemical library and identified small molecules that are non-covalent inhibitors targeting the conserved 3CL protease of SARS-CoV-2 and other coronaviruses.
Infectious #parvovirus B19 circulates in the #blood coated with active host #protease inhibitors
Source: Nature Communications, AbstractThe lack of a permissive cell culture system has limited high-resolution structures of parvovirus B19 (B19V) to virus-like particles (VLPs). In this study, we present the atomic resolution structure (2.2 Å) of authentic B19V purified from a patient blood sample. There are significant differences compared to non-infectious VLPs. Most…

#SARS-CoV-2 #Delta and #Omicron #variants resist #spike #cleavage by #human #airway trypsin-like #protease https://www.jci.org/articles/view/174304
We demonstrate that the P681R mutation in Delta and P681H mutation in the Omicron variants, adjacent to the R682 cleavage site, contributed to HAT resistance. Our study reports what we believe to be a novel soluble defense factor against SARS-CoV-2 and resistance of its actions in the Delta and Omicron variants.

The present study may contribute to gaining new insight into the roles of ISGylation-mediated anti-viral function in SARS-CoV-2 infection and may lead to the development of more potent and selective inhibitors targeted to SARS-CoV-2 nucleocapsid protein.

Monash University biologists have shed light on the intricate molecular mechanisms that are responsible for gene silencing induced by expanded repeats in an international study published today in Nature Plants.

Herein, we demonstrate that MM3122 potently inhibits viral replication in human lung epithelial cells and is also effective against the EG.5.1 variant of SARS-CoV-2.
Biochemical, structural, imaging, and functional studies reveal how the ESCRT-III protein IST1 recruits the CAPN7 cysteine protease to midbodies, where its proteolytic activity is required for efficient abscission and NoCut checkpoint arrest.
"Here, we show that #Staphylococcus aureus, a bacterial #pathogen associated with itchy skin diseases, directly activates pruriceptor sensory #neurons to drive itch. Epicutaneous S. aureus exposure causes robust itch and scratch-induced damage. By testing multiple isogenic bacterial mutants for virulence factors, we identify the S. aureus serine #protease V8 as a critical mediator in evoking spontaneous #itch and #alloknesis."
