Cellular structure without a membrane: Researcher discusses how synapses use liquids to create functional separations

The laboratory of Ege Kavalali, professor and chair of the Department of Pharmacology, published a paper in Nature Communications that determined that liquid-liquid phase separation plays a key role within the nanostructure of synapses, and that its disruption affects evoked but not spontaneous neurotransmission.

"We characterized the interaction of SNAP-25-derived peptides with Hsc70 and CSPα using a combination of biochemical and biophysical techniques, including native-PAGE, binding affinity by fluorescence anisotropy, ATPase-activity of Hsc70, and NMR. We have identified an Hsc70 binding site within SNAP-25 that is likely to represent the site used in the cell to facilitate SNARE complex formation."

"Knock out of Noelins1-3 profoundly reduced AMPARs in synapses onto excitatory and inhibitory (inter)neurons, decreased their density and clustering in #dendrites, and abolished activity-dependent #synaptic plasticity. Our results uncover an endogenous mechanism for #extracellular anchoring of AMPARs and establish Noelin-organized networks as versatile determinants of constitutive and context-dependent #neurotransmission."

Partial inhibition of class III PI3K VPS-34 ameliorates motor aging and prolongs health span

A genome-wide screen in the nematode Caenorhabditis elegans reveals potential targets to delay motor aging, including the phosphatidylinositol 3-kinase VPS-34. Genetic and pharmacological partial inhibition of VPS-34 improves neurotransmission and muscle integrity through increased PI(3)P-PI-PI(4)P conversion, ameliorating motor aging in both worms and mice.

Is #neurotransmission to #consciousness as water is to a stream? Without the water, no stream. Yet to identify a particular spot along a stream, we do so not in terms of particular physical water molecules but rather in terms of "extra-stream" landmarks. Similarly, it is in terms of relatively fixed, not flowing, elements that we "navigate" consciousness. Such entities may be, eg, synaptically weighted habits; not particles of the stream's water, but features giving rise to riffles, pools, runs.

Cortical thinning is associated with altered neurotransmission in young adults with a history of heavy drinking since adolescence

The article discusses a recent study conducted in Finland that explored the effects of #heavy #alcohol #consumption during #adolescence on #brain #development.

The study followed a group of young adults over a 10-year period, comparing those with a history of heavy drinking in adolescence to those who consumed little or no alcohol. The researchers found that the heavy-drinking group had #lower #cortical #gray #matter #thickness in several brain regions and #altered #neurotransmission compared to the control group.

Specifically, they observed increased activity in inhibitory neurotransmitters and thinner gray matter in the #frontal and #parietal lobes.

The study highlights the potential negative impact of heavy alcohol use during adolescence on brain structure and function, emphasizing the need for further research to understand the underlying mechanisms

During adolescence, the brain is particularly susceptible to the deleterious effects of alcohol use.
