The crystal structures of the 3C proteases from Coxsackievirus B3 and Coxsackievirus B4 were determined at 2.10 and 2.01 Å resolution, respectively. The structures reveal the presence of a typical chymotrypsin-like fold and a conserved catalytic triad, which are similar to those observed in 3C proteases from other enteroviruses.
RNA viruses replicate with high mutation rates, but natural selection concentrates variability in a few proteins. This study measures the effect of >40,000 non-synonymous mutations across the full proteome of coxsackievirus B3, revealing significant variation in mutational tolerance within and between viral proteins, with implications for drug target selection.
