Slow-wave sleep drives sleep-dependent renormalization of synaptic AMPA receptor levels in the hypothalamus

Sleep normalizes synaptic connections in the cortex and hippocampus that have been potentiated during the preceding wake phase. This study shows that slow-wave sleep also down-regulates AMPA receptor levels in the hypothalamus, a brain region that regulates sleep.

Cryo-ET of the SC-CA1 hippocampal connection from Eric Gouaux group. Nice confirmation that GluA2 N-terminus is closer to the pre-synapse than to the post-synapse!

Structures and analysis of three modulators that bind to #AMPAReceptor complexes including the gamma-8 subunit from Ingo Greger's lab.

Nice to see this work that was presented at the #GRC #IonChannels last year coming out, and nice to see proof of @remislp conversion to glutamate.

These compounds were found by functional screening. Considering the heterogeneity of TARP modulation that is now apparent, a strategy of rational design might never have found them.

Modulatory mechanisms of TARP γ8-selective AMPA receptor therapeutics - Nature Communications

AMPA receptors associated with TARP subunits enable the development of selective AMPA receptor drugs. Here, the authors provide cryo-EM structures of receptors bound to three TARP-γ8 selective drugs, and reveal bifunctionality of one ligand.
